Human Saitohin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Saitohin in tissue homogenates or other biological fluids. The kit uses the sandwich enzyme immunoassay technique, with an antibody specific for Saitohin pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where Saitohin is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Saitohin Antibody is added and binds the captured Saitohin. An HRP-Avidin conjugate is then introduced, and a TMB substrate solution is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. The density of yellow is proportional to the amount of Saitohin captured in each well, and concentration is calculated from the O.D. absorbance at 450 nm against a standard curve.

The assay has a sensitivity of <0.103 ng/ml and an assay range of 0.312–20 ng/ml. Detection is colourimetric and the total assay time is 4h 30m. Recovery has been demonstrated in serum (range 80–102%, average 91%, n=5), EDTA plasma (range 81–100%, average 90%, n=5), and heparin plasma (range 80–89%, average 84%, n=5). The kit is supplied with a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
