The procedure employs the sandwich enzyme immunoassay technique: an antibody specific for S100A5 is pre-coated onto a microtiter plate, standards and test samples are added to the wells, and captured S100A5 is detected using a Biotinylated Anti-S100A5 Antibody followed by HRP-Avidin conjugate and TMB substrate solution. The enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader.

Reported sensitivity is 0.085 ng/ml and the assay range is 0.312–20 ng/ml, with a total assay time of 4 hours 30 minutes.
