Human S100 Calcium Binding Protein A12 ELISA Kit

This sandwich ELISA kit from Antibodies.com enables the in vitro quantitative determination of human S100A12/CGRP (also referred to as S100 Calcium Binding Protein A12) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for the target is pre-coated onto the wells of the microplate; standards and samples are added and any target present is bound by the immobilised antibody. After washing, a Biotinylated Anti-S100A12/CGRP Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and concentration is calculated from a standard curve.

Performance specifications include a product range of 78.1-5000 pg/ml, a sensitivity of <34 pg/ml, and a total assay time of 4h 30m. Detection is colorimetric, performed on a pre-coated 12 x 8 well strip microplate, with components stored at +4°C or -20°C as instructed. Recovery has been validated in serum (average 91%, range 83%-100%, n=5), EDTA plasma (average 94%, range 90%-98%, n=5), and heparin plasma (average 95%, range 89%-102%, n=5). Linearity was demonstrated at dilutions of 1:2, 1:4, 1:8, and 1:16 across these sample types.
