Human S-Phase Kinase Associated Protein 2 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human SKP2 (S-Phase Kinase Associated Protein 2) in tissue homogenates, cell lysates, and other biological fluids. The kit uses a 96-well microtiter plate pre-coated with an antibody specific for SKP2; standards and test samples are added and SKP2 present is bound by the immobilized antibody. Following incubation and washing, a biotinylated anti-SKP2 antibody binds the captured analyte, after which an HRP-avidin conjugate and TMB substrate are added to generate a colour change that is read at 450 nm on a colorimetric microplate reader.

The quantitative sandwich assay reports a sensitivity of 0.061 ng/ml and a measuring range of 0.156-10 ng/ml, with a total assay time of 4h 30m. Spike-and-recovery validation in human samples yielded averages of 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%), and 84% in heparin plasma (range 80-89%). Linearity of dilution has been demonstrated at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma.

Reactivity is specific to human SKP2, and the kit is suited to research applications measuring SKP2 in cell lysates and tissue homogenates.
