From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human RUNX2 in tissue homogenates, cell lysates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for RUNX2. Standards and test samples are added to the wells, where RUNX2 is captured by the immobilized antibody. After washing, a Biotinylated Anti-RUNX2 Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then added to visualize the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of RUNX2 captured in each well. The concentration of RUNX2 is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.
