Human Ribonuclease L ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com, intended for the in vitro quantitative determination of human RNase L in serum, plasma, tissue homogenates or other biological fluids. The kit uses an antibody specific for RNase L pre-coated onto a 96-well microtiter plate. Standards and test samples are added, and RNase L present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-RNase L Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of RNase L captured, with concentration calculated by reading absorbance at 450 nm in a microplate reader against a standard curve.

The assay has a sensitivity of <7.3 pg/ml and a range of 15.6-1000 pg/ml, with a total run time of 4h 30m using colorimetric detection on a microplate reader. Reported average recovery is 91% in serum (range 80-102%), 91% in EDTA plasma (range 81-101%), and 84% in heparin plasma (range 80-89%), each across n=5 samples. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions: serum recovery 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma recovery 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma recovery 84-95%, 92-105%, 82-105% and 89-91% (all n=5). Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.
