Human Ribonuclease III, Nuclear ELISA Kit

From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of human Drosha (Ribonuclease III, nuclear) in tissue homogenates, cell lysates or other biological fluids. The assay employs a 96-well microplate (12 x 8 well strips) pre-coated with an antibody specific to the target. Standards and samples are added to the wells, followed by a Biotinylated Anti-Drosha Antibody and an HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of stop solution; absorbance is read at 450 nm in a colorimetric microplate reader, with colour intensity proportional to Drosha concentration. The stated assay range is 0.312–20 ng/ml and sensitivity is <0.126 ng/ml, with a total assay time of 4h 30m.

Reported mean recovery from serum (n=5) is 91% (range 80%–102%), from EDTA plasma (n=5) is 91% (range 81%–101%), and from heparin plasma (n=5) is 84% (range 80%–89%). Dilution-linearity values across 1:2, 1:4, 1:8 and 1:16 dilutions are also provided for each of the three sample types.
