From Antibodies.com, this competitive ELISA is designed for the in vitro quantitative determination of human Relaxin in serum, plasma or other biological fluids, read on a colorimetric microplate reader. The assay employs the competitive enzyme immunoassay technique, in which Relaxin present in the samples or standards competes with Relaxin antigen pre-coated on a 96-well microplate for binding sites on a biotinylated anti-Relaxin antibody. Following incubation and washing to remove unbound antibody, an HRP-avidin conjugate is added; TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow after addition of acidic stop solution. The intensity of the yellow colour is inversely proportional to the amount of Relaxin in each sample or standard, and the concentration is calculated by reading the absorbance at 450 nm and referring to the standard curve.

The assay covers a working range of 31.2–2000 pg/ml with a reported sensitivity of 9.66 pg/ml, and the protocol is completed in approximately 2 hours 30 minutes. Kit components (pre-coated microplate, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers) are stored between +4°C and −20°C as instructed, and the kit should not be used past its expiration date.

Validation studies report recovery values for serum (range 80–102%, mean 91%, n=5), EDTA plasma (range 81–100%, mean 90%, n=5) and heparin plasma (range 80–89%, mean 84%, n=5), with linearity upon dilution of the same sample types evaluated across 1:2 to 1:16 dilutions.
