Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human Receptor Activator of Nuclear Factor Kappa B Ligand (RANKL) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with a capture antibody specific for RANKL. Standards and test samples are added to the wells so that RANKL present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-RANKL Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of RANKL captured per well, and the concentration is calculated from the O.D. absorbance read at 450 nm in a colorimetric microplate reader.

The kit is suitable for serum, plasma, cell lysates, tissue homogenates and cell culture supernates, and the platform is a pre-coated microplate of 12 x 8 well strips. Reported assay performance includes a sensitivity of 0.056 ng/ml and a product range of 0.156-10 ng/ml, with an assay time of approximately 4h 30m. Kit components are to be stored at +4°C or -20°C as instructed in the protocol booklet, and components must not be used past the expiration date. The product is intended for research use only and is not intended for diagnostic or therapeutic use.
