Human Ras Homolog Gene Family, Member A ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human RhoA in tissue homogenates, cell lysates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for RhoA. Standards and test samples are added to the wells, where RhoA is captured by the immobilised antibody. A Biotinylated Anti-RhoA Antibody is then added, followed by HRP-Avidin conjugate and TMB substrate solution. The reaction produces a blue-coloured product that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of RhoA captured. The concentration of RhoA is calculated by reading O.D. absorbance at 450 nm in a microplate reader and referring to a standard curve.

The kit has a stated assay range of 0.156–10 ng/ml and a sensitivity of 0.057 ng/ml. The colourimetric quantitative assay takes 4h 30m to perform. Storage is at +4°C or -20°C as instructed, with a warning not to use past the expiration date.

Sample recovery has been characterised for serum, EDTA plasma, and heparin plasma. Reported recovery averages across the tested sample types were 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%), and 84% for heparin plasma (range 80%–89%).
