Human Pyruvate Kinase, Muscle ELISA Kit

Manufactured by Antibodies.com for the in vitro quantitative determination of human Pyruvate Kinase Isozyme M1 in serum, plasma, tissue homogenates or other biological fluids, this kit uses a sandwich enzyme immunoassay format with reactivity for human samples.

The assay employs a 96-well microplate pre-coated with an antibody specific for Pyruvate Kinase Isozyme M1, configured as 12 x 8 well strips. Standards and test samples are added to the wells; captured target is bound by a Biotinylated Anti-Pyruvate Kinase Isozyme M1 Antibody and detected with an HRP-Avidin conjugate. TMB substrate is converted to a blue product that becomes yellow upon addition of acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader. The standard curve is used to calculate concentration. The product range is 78.1–5000 pg/ml with a sensitivity of <29 pg/ml, and the total assay time is 4h 30m. Supplied components include lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 µl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips), with storage at +4°C or -20°C as instructed.

Reported recovery values are: serum average 91% (range 80–102%, n=5), EDTA plasma average 90% (range 81–100%, n=5) and heparin plasma average 84% (range 80–89%, n=5). Dilution linearity data are provided for 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma.
