Human Pyruvate Dehydrogenase Beta ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Pyruvate Dehydrogenase Beta (PDHB) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate and follows a standard sandwich immunoassay workflow: standards and samples are added to the wells, captured PDHB is detected using a Biotinylated Anti-PDHB Antibody followed by an HRP-Avidin conjugate, and signal is developed with TMB substrate. The colour change is read at 450 nm using a microplate reader, and PDHB concentration is calculated by reference to a standard curve.

The standard curve covers a measuring range of 1.56-100 U/L, with a stated sensitivity of <1.2 U/L. Suitable sample types include serum, EDTA plasma, heparin plasma, cell culture supernatant, tissue homogenates and cell lysates. Spike-and-recovery testing gave an average of 91% (range 80%-102%, n=5) for serum, 90% (range 81%-100%, n=5) for EDTA plasma and 84% (range 80%-89%, n=5) for heparin plasma. Dilution linearity was demonstrated across 1:2 to 1:16 dilutions for each of these sample matrices. Total assay time is approximately 4 hours 30 minutes. Provided components include a pre-coated microplate arranged as 12 x 8 well strips, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, to be stored at +4°C or -20°C as instructed.
