Human Pyruvate Carboxylase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Pyruvate Carboxylase (PCB) in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated antibody specific for PCB immobilised on a microplate. Standards and samples are added to the wells; after incubation and washing, a Biotinylated Anti-PCB Antibody binds the captured analyte, followed by HRP-Avidin conjugate. TMB substrate is added to visualise the enzymatic reaction, yielding a blue colour that turns yellow upon addition of stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, with PCB concentration calculated against the standard curve.

The working range is 1.56–100 ng/ml with a sensitivity of 0.63 ng/ml, and the total assay time is 4 hours 30 minutes. Recovery has been demonstrated in serum (80%–102%, average 91%), EDTA plasma (81%–99%, average 90%) and heparin plasma (80%–89%, average 84%), with dilution linearity assessed at 1:2, 1:4, 1:8 and 1:16 across these matrices. The platform is a pre-coated microplate arranged as 12 × 8 well strips, and kit components are stored at +4°C or −20°C as instructed.
