Human Purinergic Receptor P2Y, G Protein Coupled 14 ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human P2Y14 in tissue homogenates, cell lysates and other biological fluids.

The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for P2Y14 pre-coated onto a 96-well microtiter plate (12 x 8 well strips). Standards and test samples are added and P2Y14 present in each sample is bound by the immobilised antibody. After washing, a Biotinylated Anti-P2Y14 Antibody is added, followed by HRP-Avidin conjugate. Following further incubation and washing, TMB substrate solution is added to visualise the HRP enzymatic reaction; the resulting blue product changes to yellow upon addition of acidic stop solution. The density of yellow, read as absorbance at 450 nm on a colorimetric microplate reader, is proportional to the amount of P2Y14 captured in each well.

Performance figures include a sensitivity of 0.053 ng/ml and an assay range of 0.156-10 ng/ml. Recovery in serum (n=5) was 83-100% with an average of 91%; in EDTA plasma (n=5) 90-98% with an average of 94%; and in heparin plasma (n=5) 89-102% with an average of 95%. Linearity was assessed at dilutions of 1:2, 1:4, 1:8 and 1:16. Total assay time is 4 h 30 min. Kit components include a pre-coated microplate, lyophilised standard, detection reagents, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored according to instructions at +4°C or -20°C.
