Human Protoporphyrinogen Oxidase ELISA Kit

From Antibodies.com, this Human PPOX ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Protoporphyrinogen Oxidase (PPOX) in tissue homogenates or other biological fluids. It is reactive with human samples and runs on a pre-coated microplate platform with colorimetric detection read at 450 nm on a colorimetric microplate reader.

An antibody specific for PPOX is pre-coated on the plate. Standards and samples are added and the PPOX present is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-PPOX Antibody is added, then an HRP-Avidin conjugate, and finally TMB substrate solution. The HRP enzymatic reaction produces a blue-coloured product that changes to yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of PPOX captured in each well, and concentration is calculated from the standard curve. Sensitivity is <0.053 ng/ml and the assay range is 0.156–10 ng/ml, with a total assay time of 4h 30m. Recovery in serum averaged 91% (range 80–102%), in EDTA plasma averaged 90% (range 81–100%), and in heparin plasma averaged 84% (range 80–89%). Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
