Human Protocadherin Beta 16 ELISA Kit

This sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human PCDHB16 (Protocadherin Beta 16) in tissue homogenates or other biological fluids. The assay uses the sandwich immunoassay format on a pre-coated 96-well microplate: standards and test samples are added to the wells, and PCDHB16 present in each sample is bound by the immobilised capture antibody. After washing, a biotinylated anti-PCDHB16 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The concentration of PCDHB16 is calculated from the optical density at 450 nm in a microplate reader by reference to a standard curve.

The kit has a sensitivity of <0.055 ng/ml and a quantitative range of 0.156–10 ng/ml. Total assay time is 4 h 30 min and detection is colourimetric. Reported average spike-and-recovery values for serum, EDTA plasma, and heparin plasma (n=5 per matrix) are 91% (range 83–100%), 94% (range 90–98%), and 95% (range 89–102%), respectively.
