The technique uses an antibody specific for Myelin PLP pre-coated onto a microplate; standards and test samples are added to the wells and bound analyte is detected with a Biotinylated Anti-Myelin PLP Antibody and HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate solution and the absorbance is read at 450 nm using a colorimetric microplate reader.

The kit reports a sensitivity of 0.053 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4 hours 30 minutes. Average recoveries (n=5) for the listed sample types are 91% in serum, 90% in EDTA plasma and 84% in heparin plasma.
