Human Proteoglycan 4 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of human Lubricin/MSF, also referred to as Proteoglycan 4, in serum, plasma, synovial fluid or other biological fluids. The assay is configured on a pre-coated 96-well microplate supplied as 12 x 8 well strips, with detection carried out on a colorimetric microplate reader.

An antibody specific for Lubricin/MSF is immobilised on the plate; standards and test samples are added, followed by a Biotinylated Anti-Lubricin/MSF Antibody and an HRP-Avidin conjugate. After incubation and washing, TMB substrate is added, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm and the concentration is calculated against the standard curve. Reported sensitivity is <0.54 ng/ml, the assay range is 1.25–80 ng/ml, and the total assay time is 4h 30m.

Validation data are provided for serum, EDTA plasma and heparin plasma. Spike-and-recovery results are n=5, range 80%–102%, average 91% for serum; n=5, range 81%–100%, average 90% for EDTA plasma; and n=5, range 80%–89%, average 84% for heparin plasma. Dilution linearity is reported for the same three sample types across 1:2, 1:4, 1:8 and 1:16 dilutions. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
