Human Protein Tyrosine Phosphatase Type IVA 3 ELISA Kit

The assay is reactive with human samples and uses a pre-coated microplate with colorimetric detection read at 450 nm on a standard microplate reader.

Performance specifications include an assay range of 0.312–20 ng/ml and a sensitivity of <0.118 ng/ml, with a total assay time of 4h 30m. Validation across sample matrices (n=5) showed average recoveries of 91% in serum (range 80–102%), 91% in EDTA plasma (range 81–101%), and 84% in heparin plasma (range 80–89%). Dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 across serum, EDTA plasma, and heparin plasma samples.

The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for PTP4A3/PRL-R pre-coated on the plate. Standards and samples are added, followed by a biotinylated anti-PTP4A3/PRL-R antibody, HRP-avidin conjugate, and TMB substrate. The HRP-catalysed reaction produces a blue product that turns yellow upon addition of acidic stop solution, with absorbance proportional to PTP4A3/PRL-R concentration.
