From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human DEP1 (Protein Tyrosine Phosphatase Receptor Type J) in tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using a 96-well microtiter plate pre-coated with an antibody specific for DEP1. Standards and test samples are added to the wells and any DEP1 present is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-DEP1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution.

The product range is 0.312–20 ng/ml with a sensitivity of <0.117 ng/ml. Absorbance is read at 450 nm in a colorimetric microplate reader, and total assay time is 4 hours 30 minutes. The platform consists of pre-coated 12 x 8 well strips. Recovery has been validated in serum (range 80%–102%, average 91%, n=5), EDTA plasma (range 81%–100%, average 90%, n=5) and heparin plasma (range 80%–89%, average 84%, n=5). Kit components include a pre-coated 96-well microplate, lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Components should be stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date.
