From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human PTP1B in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique: an antibody specific for PTP1B is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips, and standards and test samples are added so that PTP1B is captured by the immobilised antibody. After washing, a Biotinylated Anti-PTP1B Antibody binds the captured PTP1B, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of an acidic stop solution; the absorbance is read at 450 nm in a microplate reader and the concentration of PTP1B is calculated by reference to a standard curve.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.054 ng/ml, and the total assay time is 4h 30m. Detection is colourimetric using a microplate reader. Recovery and dilution linearity data are provided for serum, EDTA plasma, and heparin plasma samples. Kit components are stored at +4°C or –20°C as instructed and the kit should not be used past its expiration date.
