A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Protein Kinase N1 (PKN1) in tissue homogenates, cell lysates, or other biological fluids. The kit is reactive with human samples and uses a quantitative sandwich format with colorimetric detection read at 450 nm on a standard microplate reader.

According to the manufacturer, the assay has a working range of 0.312–20 ng/ml, a sensitivity of <0.117 ng/ml, and a total assay time of approximately 4 hours 30 minutes. A capture antibody specific for PKN1 is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Bound PKN1 is detected using a biotinylated anti-PKN1 antibody, an HRP-avidin conjugate, and TMB substrate; the enzymatic reaction is stopped with acidic stop solution and the optical density is read against a standard curve.

Reported average recovery values from the manufacturer (n=5 per matrix) are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Linearity data across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5) are reported as 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. The kit contains a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, with each component stored at +4°C or –20°C as indicated.
