Human Protein Kinase C Zeta ELISA Kit

Distributed by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Protein Kinase C Zeta (PKC zeta) in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) bearing an antibody specific for PKC zeta. Standards and test samples are added to the wells, followed by a Biotinylated Anti-PKC zeta Antibody, an HRP-Avidin conjugate, and a TMB substrate solution. The HRP-catalysed reaction yields a blue product that turns yellow upon addition of the acidic stop solution, with the intensity of yellow proportional to the amount of PKC zeta captured in each well. Absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated from the standard curve.

Reported sensitivity is <0.112 ng/ml and the assay range spans 0.312-20 ng/ml. The complete protocol takes approximately 4h 30m. Sample recovery has been characterised: in serum, n=5, the average is 91% across a range of 80-102%; in EDTA plasma, n=5, the average is 90% across a range of 81-100%; in heparin plasma, n=5, the average is 84% across a range of 80-89%. Linearity of dilution has also been reported from 1:2 through 1:16 for serum, EDTA plasma, and heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
