Human Protein Kinase B Beta ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human AKT2 (Protein Kinase B Beta) in tissue homogenates, cell lysates, and other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for AKT2 that has been pre-coated onto a 96-well microtiter plate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where AKT2 is captured by the immobilized antibody. After washing, a Biotinylated Anti-AKT2 Antibody binds the captured target, and an HRP-Avidin conjugate is added. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour, read as O.D. absorbance at 450nm in a microplate reader, is proportional to the amount of AKT2 present and is quantified against a standard curve.

The reported assay sensitivity is <0.132 ng/ml, with a working range of 0.312–20 ng/ml. Recovery performance in serum averaged 91% (range 80%–102%, n=5), in EDTA plasma averaged 90% (range 81%–100%, n=5), and in heparin plasma averaged 84% (range 80%–89%, n=5).
