Human Protein Kinase, AMP Activated Alpha 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human AMPK alpha 1 in tissue homogenates, cell lysates, or other biological fluids. The kit employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with absorbance read at 450 nm using a colorimetric microplate reader. The assay has a reported sensitivity of 0.062 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of approximately 4 hours 30 minutes.

Biotinylated Anti-AMPK alpha 1 Antibody is used for detection, followed by an HRP-Avidin conjugate and TMB substrate; the enzymatic reaction produces a blue-coloured product that turns yellow upon addition of the acidic stop solution, with the intensity of the yellow colour proportional to AMPK alpha 1 concentration. Standards and test samples are added to the pre-coated wells, and following incubation and washing steps the captured analyte is detected sequentially. Kit components are stored at +4°C or -20°C as instructed, and the product is intended for research use only.

Recovery has been validated in human serum (average 91%, range 80–102% across n=5), EDTA plasma (average 91%, range 81–101%), and heparin plasma (average 84%, range 80–89%). Linearity has been demonstrated across dilution factors of 1:2, 1:4, 1:8, and 1:16 in serum (ranges 70–107%, 89–93%, 86–104%, 80–103% respectively), EDTA plasma (95–100%, 85–110%, 81–99%, 90–102%), and heparin plasma (77–104%, 88–95%, 94–104%, 80–107%).
