Human Protein Disulfide Isomerase A4 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of human ERp72 (Protein Disulfide Isomerase A4) in serum, plasma, tissue homogenates or other biological fluids. Distributed under the Antibodies.com brand, the kit uses an antibody specific for ERp72 pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added, followed by a Biotinylated Anti-ERp72 Antibody, an HRP-Avidin conjugate and TMB substrate; the resulting colour change is read at 450 nm in a colorimetric microplate reader and the ERp72 concentration is calculated from the standard curve.

The stated sensitivity is <0.66 ng/ml and the assay range is 1.56–100 ng/ml, with a total assay time of 4 hours 30 minutes. Reported recovery values (n=5) are 80%–102% (average 91%) in serum, 81%–100% (average 90%) in EDTA plasma and 80%–89% (average 84%) in heparin plasma, and linearity data are given across 1:2, 1:4, 1:8 and 1:16 dilutions for each matrix. Kit components comprise a pre-coated 12 x 8 well strip microplate, 2 vials of lyophilised standard, detection solutions A and B (120 µl each), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers, with storage at +4°C or -20°C as indicated per component.
