Human Protein Disulfide Isomerase A3 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of human ERp57 (also known as Protein Disulfide Isomerase A3) in tissue homogenates, cell lysates, and other biological fluids. The kit employs a 96-well microtiter plate pre-coated with a capture antibody specific for ERp57, to which standards and test samples are added; ERp57 present in each sample binds to the immobilized antibody.

Following incubation and washing, a Biotinylated Anti-ERp57 Antibody is added, which binds the captured analyte. An HRP-Avidin conjugate is then introduced, and the enzymatic reaction is visualised using TMB substrate, producing a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader. The assay demonstrates a sensitivity of <0.055 ng/ml and a quantitative range of 0.156–10 ng/ml, with a total run time of 4 hours 30 minutes. Reported recovery data include: Serum (n=5) 80–102% (average 91%); EDTA Plasma (n=5) 81–100% (average 90%); Heparin Plasma (n=5) 80–89% (average 84%).
