Human Proteasome 26S Subunit, Non ATPase 10 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Proteasome 26S Subunit, Non ATPase 10 (referred to in the manufacturer's protocol as Gankyrin) in tissue homogenates or other biological fluids. The assay employs a pre-coated microplate on which an antibody specific for Gankyrin has been immobilised; standards and test samples are added and the target is captured by the coated antibody. After washing, a biotinylated anti-Gankyrin detection antibody is added, followed by HRP-avidin conjugate. The HRP enzymatic reaction is visualised using TMB substrate solution, producing a blue product that turns yellow upon addition of stop solution; the absorbance at 450 nm, read on a colorimetric microplate reader, is proportional to the amount of Gankyrin present and is quantified by reference to a standard curve.

The kit has an assay range of 0.156–10 ng/ml and a sensitivity of <0.059 ng/ml. Detection is colorimetric, the assay type is sandwich (quantitative), and the total assay time is 4 hours 30 minutes. Reported average recovery values across spiked samples are 91% in serum (n=5, range 83–100%), 94% in EDTA plasma (n=5, range 90–98%) and 95% in heparin plasma (n=5, range 89–102%). Linearity has been demonstrated at sample dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma.
