From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of human PAR1/Thrombin Receptor in tissue homogenates, cell lysates and other biological fluids [S1]. Reactivity is specified as Human [C0][S1].

The assay employs a pre-coated 96-well microtiter plate, with standards and samples added so that PAR1/Thrombin Receptor is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-PAR1/Thrombin Receptor antibody is added, then an HRP-avidin conjugate. The signal is developed with TMB substrate and an acidic stop solution, producing a yellow product read at 450 nm on a colorimetric microplate reader against a standard curve [S1]. The stated product range is 0.156–10 ng/ml with a sensitivity of <0.059 ng/ml, and the total assay time is 4 hours 30 minutes [S1].

Reported recovery in undiluted samples (n=5) is: Serum 80–102% (average 91%); EDTA Plasma 81–100% (average 90%); Heparin Plasma 80–89% (average 84%). Linearity across 1:2 to 1:16 dilutions (n=5) is also reported for serum, EDTA plasma, and heparin plasma [S1]. Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and plate sealers [S1].
