Human Protamine 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Protamine 2 in tissue homogenates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for Protamine 2. Standards and test samples are added to the wells and Protamine 2 present in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-Protamine 2 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the enzymatic reaction; the colour changes from blue to yellow upon addition of acidic stop solution, with the density of yellow proportional to the amount of Protamine 2 captured. Absorbance is read at 450 nm in a microplate reader and the concentration calculated by reference to the standard curve.

The assay has a product range of 0.156–10 ng/ml and a sensitivity of 0.057 ng/ml. The platform is a pre-coated microplate comprising 12 x 8 well strips, with a total assay time of 4h 30m. Manufacturer-reported recovery values are an average of 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity ranges are reported for serum, EDTA plasma and heparin plasma across dilutions of 1:2, 1:4, 1:8 and 1:16. Kit components include a pre-coated 96 well microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
