Human Proprotein Convertase Subtilisin/Kexin Type 1 ELISA Kit

This sandwich ELISA kit from Antibodies.com is intended for the in vitro quantitative determination of human PC1/3 (Proprotein Convertase Subtilisin/Kexin Type 1) in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for PC1/3 is pre-coated onto a microplate, standards and test samples are added and bound, the wells are washed, and a Biotinylated Anti-PC1/3 Antibody is then incubated in the wells. An HRP-Avidin conjugate is added, and TMB substrate solution is used to visualise the HRP enzymatic reaction. The blue product turns yellow upon addition of acidic stop solution, and the absorbance is read at 450 nm in a microplate reader with the concentration calculated against a standard curve.

The kit has a reported sensitivity of 0.101 ng/ml and an assay range of 0.312-20 ng/ml. Recovery data for serum (n=5) shows a range of 80%-102% with an average of 91%; EDTA plasma (n=5) shows 81%-100% with an average of 90%; heparin plasma (n=5) shows 80%-89% with an average of 84%. Dilution linearity for serum (n=5) at 1:2, 1:4, 1:8 and 1:16 gave ranges of 87-91%, 87-107%, 74-101% and 92-97% respectively. EDTA plasma (n=5) gave 90-105%, 84-101%, 90-101% and 79-108%, while heparin plasma (n=5) gave 84-95%, 92-105%, 82-105% and 89-91%. The assay takes 4h 30m, and components should be stored at +4°C or -20°C as instructed. The kit must not be used past its expiration date.
