Human Prolyl-4-Hydroxylase Alpha Polypeptide II ELISA Kit

The Antibodies.com Human P4HA2 ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Prolyl 4 Hydroxylase Alpha Polypeptide II (P4HA2) in tissue homogenates or other biological fluids. Intended for research use only, it is not for diagnostic or therapeutic applications.

The assay employs a pre-coated antibody specific for P4HA2 on a microplate. Standards and samples are added; captured P4HA2 is detected using a Biotinylated Anti-P4HA2 Antibody and an HRP-Avidin conjugate, with TMB substrate producing a colour change from blue to yellow after addition of the acidic stop solution. Absorbance is read at 450nm in a colorimetric microplate reader, and P4HA2 concentration is determined against the standard curve.

Sensitivity is 0.063 ng/ml, the assay range is 0.156-10 ng/ml, and the total assay time is 4h 30m. Recovery has been validated for serum (80%-102%, average 91%), EDTA plasma (81%-100%, average 90%) and heparin plasma (80%-89%, average 84%). Linearity has been demonstrated across sample dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Reagents are kept at +4°C or -20°C as instructed and should not be used past the expiration date. Serum, plasma, cell culture supernatant, tissue homogenates, cell lysates and other biological fluids may be used; samples can be stored at -20°C for up to 1 month or -80°C for up to 6 months, avoiding repeated freeze/thaw cycles.
