The assay employs a pre-coated microplate onto which an antibody specific for PCNA has been immobilised; standards and test samples are added, followed by a Biotinylated Anti-PCNA Antibody and an HRP-Avidin conjugate. Signal is visualised using a TMB substrate solution that produces a yellow product upon addition of the acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader. The density of yellow is proportional to the amount of PCNA captured in each well, and concentration is calculated by reference to the standard curve.

Reported performance specifications include a measuring range of 0.312–20 ng/ml and a sensitivity of <0.123 ng/ml. The complete assay requires approximately 4h 30m, and kit components are stored at +4°C or -20°C as indicated on the component label. Validated recovery in human serum, EDTA plasma and heparin plasma averages 84–91%, with dilution linearity demonstrated across 1:2 to 1:16 dilutions in each matrix.
