Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Prolactin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for Prolactin is immobilised on the plate; standards and samples are added, captured Prolactin is detected using a Biotinylated Anti-Prolactin Antibody followed by an HRP-Avidin conjugate, and signal is generated with TMB substrate. Absorbance is read at 450 nm on a colorimetric microplate reader, with the intensity of the yellow product proportional to the amount of Prolactin captured in each well.

The measuring range is 1.56–100 ng/ml and the reported sensitivity is <0.56 ng/ml, with a total assay time of 4h 30m. In spike-and-recovery testing with n=5 samples per matrix, serum showed an average recovery of 91% over a range of 80%–102%; EDTA plasma averaged 90% (range 81%–100%); and Heparin plasma averaged 84% (range 80%–89%). Dilution linearity at 1:2, 1:4, 1:8, and 1:16 gave serum recoveries of 87–91%, 87–107%, 74–101%, and 92–97%; EDTA plasma 90–105%, 84–101%, 90–101%, and 79–108%; and Heparin plasma 84–95%, 92–105%, 82–105%, and 89–91%.

Kit components include a pre-coated 96-well microplate, a lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
