Human Prokineticin 2 ELISA Kit

A sandwich ELISA kit from Antibodies.com for the in vitro quantitative determination of human Prokineticin 2 (PK2) in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate (12 × 8 well strips) with an immobilised antibody specific for Prokineticin 2/PK2, a biotinylated anti-Prokineticin 2/PK2 detection antibody, HRP-avidin conjugate and TMB substrate. The reaction is stopped with acidic stop solution and absorbance is read at 450 nm on a colourmetric microplate reader; the concentration of analyte is calculated against a standard curve.

According to the manufacturer's specification, the working assay range is 0.312–20 ng/ml and sensitivity is <0.084 ng/ml. Total assay time is 4 h 30 min. Spike-and-recovery values (n=5) were 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma and 80–89% (average 84%) for heparin plasma. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87–91% / 87–107% / 74–101% / 92–97% for serum, 90–105% / 84–101% / 90–101% / 79–108% for EDTA plasma and 84–95% / 92–105% / 82–105% / 89–91% for heparin plasma (n=5 per matrix).

Components supplied include the pre-coated microplate, lyophilised standard (2 vials), detection solutions A and B (120 µl each), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers (5 adhesive strips). Kit components should be stored at +4 °C or −20 °C as instructed and not used past the expiration date.
