The assay employs a pre-coated microplate to which standards and samples are added; captured PDCD6/ALG-2 is detected using a biotinylated anti-PDCD6/ALG-2 antibody, HRP-Avidin conjugate, and TMB substrate. The reaction produces a yellow product measured by absorbance at 450 nm on a colorimetric microplate reader, with PDCD6/ALG-2 concentration calculated against a standard curve. Total assay time is 4h 30m.

Performance characteristics quoted from the manufacturer include a sensitivity of <0.118 ng/ml and an assay range of 0.312-20 ng/ml. Spike-and-recovery values in serum (n=5) were 80-102% with an average of 91%; in EDTA plasma (n=5) 81-100% with an average of 90%; and in heparin plasma (n=5) 80-89% with an average of 84%. Dilution linearity across 1:2 to 1:16 dilutions (n=5) was reported as 87-91% to 92-97% for serum, 90-105% to 79-108% for EDTA plasma, and 84-95% to 89-91% for heparin plasma.
