Human Procollagen C Proteinase Enhancer 1 ELISA Kit

Sandwich ELISA kit from Antibodies.com designed for the in vitro quantitative determination of human Procollagen C Proteinase Enhancer 1 (PCOLCE) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is reactive with human samples and uses a pre-coated 96-well microplate platform (12 x 8 well strips), with detection carried out on a colorimetric microplate reader.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for PCOLCE is pre-coated onto the plate; standards and test samples are added and the PCOLCE present is captured by the immobilised antibody. After washing, a Biotinylated Anti-PCOLCE Antibody binds the captured PCOLCE, followed by incubation with HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow upon addition of the stop solution. The density of yellow is proportional to the amount of PCOLCE captured, and concentration is calculated by reading absorbance at 450 nm and referring to the standard curve. Total assay time is 4h 30m, and components are stored at +4°C or -20°C as instructed; the kit should not be used past its expiration date.

Reported product range is 0.625–40 ng/ml with a sensitivity of <0.238 ng/ml. Sample recovery (n=5) was 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma, and 80–89% (average 84%) for heparin plasma. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5) was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
