Human Prekallikrein ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Prekallikrein in plasma. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for Prekallikrein. Standards and samples are added to the wells, where Prekallikrein binds to the immobilised antibody. After incubation and washing, a Biotinylated Anti-Prekallikrein Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of Prekallikrein captured, and concentration is calculated from the O.D. absorbance at 450 nm read on a colorimetric microplate reader against the standard curve.

According to the manufacturer, the assay has a stated range of 1.56-100 ng/ml and a sensitivity of <0.64 ng/ml, with a total assay time of 4h 30m. Reported recovery values (n=5) are 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
