Human Pregnane X Receptor ELISA Kit

From Antibodies.com, this Sandwich ELISA is designed for the in vitro quantitative determination of human Pregnane X Receptor (PXR) in tissue homogenates, cell lysates or other biological fluids. The assay is reactive with human samples and uses colorimetric detection on a microplate reader platform.

The kit employs a sandwich enzyme immunoassay technique run on a pre-coated 96-well microtiter plate. Standards and test samples are added to the wells, where PXR is captured by an immobilised specific antibody. After washing, a Biotinylated Anti-PXR Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of stop solution. The density of yellow is proportional to the amount of PXR captured, and the concentration is calculated from the O.D. absorbance read at 450 nm. Total assay time is 4 hours 30 minutes.

Reported performance characteristics include a sensitivity of <0.126 ng/ml and an assay range of 0.312–20 ng/ml. Spike-and-recovery testing (n=5) yielded average recoveries of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity from 1:2 to 1:16 (n=5) gave serum recoveries of 87–91%, 87–107%, 74–101% and 92–97% respectively, with EDTA plasma recoveries of 90–105%, 84–101%, 90–101% and 79–108%, and heparin plasma recoveries of 84–95%, 92–105%, 82–105% and 89–91%. Kit components should be stored at +4°C or −20°C as instructed and the product must not be used past its expiration date.
