Description

Manufactured by Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human PRAME (Preferentially Expressed Antigen In Melanoma) in tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated microplate and is read on a colorimetric microplate reader at 450 nm.

In the workflow, standards and test samples are added to the wells and PRAME is captured by an immobilized specific antibody. After washing, a Biotinylated Anti-PRAME Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualize the reaction, producing a blue product that changes to yellow upon addition of acidic stop solution; the intensity of the yellow colour is proportional to the amount of PRAME captured. The quantitative detection range is 0.312–20 ng/ml with a sensitivity of <0.105 ng/ml, and total assay time is 4 h 30 min.

Recovery performance has been characterised in three matrices with an average of 91% for serum (n=5, range 80%–102%), 90% for EDTA plasma (n=5, range 81%–100%) and 84% for heparin plasma (n=5, range 80%–89%). Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
