From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (sELISA) is designed for the in vitro quantitative determination of human KCNN2/SK2, also referred to as Potassium Intermediate Small Conductance Calcium Activated Channel Subfamily N, Member 2. It is intended for tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and reactivity is confirmed for human samples.

The assay uses a pre-coated microplate supplied as 12 x 8 well strips and is read on a colorimetric microplate reader at 450 nm, with a total assay time of 4h 30m. The reported product range is 0.156–10 ng/ml and sensitivity is stated as <0.081 ng/ml. Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.

In the sandwich immunoassay format, standards and samples are captured by an antibody immobilised on the plate and then detected using a biotinylated anti-KCNN2/SK2 antibody followed by an HRP-avidin conjugate. TMB substrate is added and the enzymatic reaction is stopped with an acidic solution, producing a yellow colour whose absorbance is proportional to the amount of KCNN2/SK2 present in each well, allowing the concentration to be calculated from a standard curve.
