Human Pleiotrophin ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of human pleiotrophin (PTN) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses a pre-coated microplate to which an antibody specific for PTN is immobilised. Standards and test samples are added; PTN present in each sample is bound by the immobilised antibody. After washing, a biotinylated anti-PTN antibody is added, followed by an HRP-avidin conjugate. The signal is visualised with TMB substrate, yielding a blue product that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and PTN concentration is calculated from the standard curve. Total assay time is 4 hours 30 minutes.

The working range is 15.6-1000 pg/ml with a sensitivity of <7.1 pg/ml. Reported recovery (n=5) is 80-102% (average 91%) in serum, 81-100% (average 90%) in EDTA plasma and 80-89% (average 84%) in heparin plasma. Kit components are stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date.
