From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Platelet Derived Growth Factor Receptor Beta (PDGFR beta) in tissue homogenates, cell lysates or other biological fluids. The kit is validated for human reactivity and detection is performed on a colorimetric microplate reader.

The assay uses a 96-well microtiter plate pre-coated with an antibody specific for PDGFR beta. Standards and samples are added to the wells where PDGFR beta binds the immobilized capture antibody. After washing, a Biotinylated Anti-PDGFR beta Antibody is added and binds the captured target. HRP-Avidin conjugate is added and, following further incubation and washing, TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm and PDGFR beta concentration is calculated from the standard curve. Total assay time is 4 hours 30 minutes.

Reported performance characteristics include a sensitivity of less than 0.063 ng/ml and an assay range of 0.156–10 ng/ml. Spike-and-recovery testing in serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5) gave recovery ranges of 80–102% (average 91%), 81–101% (average 91%) and 80–89% (average 84%) respectively. The kit is supplied with a pre-coated 96-well microplate (12 x 8 well strips), two vials of lyophilized standard, Detection Solution A and Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and five adhesive plate sealers, with components stored at +4°C or -20°C as instructed in the protocol.
