Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Platelet Derived Growth Factor BB (PDGFBB) in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate onto which an antibody specific for PDGFBB has been immobilised. Standards and test samples are added to the wells, and any PDGFBB present is captured by the immobilised antibody. After washing, a Biotinylated Anti-PDGFBB Antibody is added and binds the captured PDGFBB, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of PDGFBB captured in each well, and concentration is calculated from the O.D. absorbance read at 450 nm in a microplate reader against the standard curve.

Reported sensitivity is 13.4 pg/ml with an assay range of 31.2–2000 pg/ml. The total assay time is 4 hours 30 minutes. Recovery has been evaluated in serum (n=5, 80%–102%, average 91%), EDTA plasma (n=5, 81%–101%, average 91%) and heparin plasma (n=5, 80%–89%, average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is reported as 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components should be stored at +4°C or –20°C as instructed, and the kit must not be used past its expiration date.

The assay is intended for use with a colorimetric microplate reader and is suitable for serum and plasma samples.
