Human Platelet Derived Growth Factor AA ELISA Kit

This sandwich ELISA kit from Antibodies.com is intended for the in vitro quantitative determination of human Platelet Derived Growth Factor AA (PDGFAA) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. It is supplied on a pre-coated microplate (12 x 8 well strips) and produces a colourimetric readout measured on a microplate reader at 450 nm.

According to the manufacturer, the assay has a standard range of 0.156–10 ng/ml and a sensitivity of <0.055 ng/ml. The complete protocol can be carried out in 4h 30m. Detection relies on a biotinylated anti-PDGFAA antibody followed by an HRP-avidin conjugate, with TMB substrate generating a yellow product after addition of acidic stop solution; the absorbance is proportional to the PDGFAA concentration in each well and is quantified against the standard curve.

Spike-and-recovery has been evaluated on five replicates of serum, EDTA plasma and heparin plasma, with mean recoveries of 91%, 91% and 84% respectively and an overall observed range of 80–102%. Dilution linearity from 1:2 to 1:16 (n=5) gives recoveries within 74–108% across the tested matrices. Kit components are stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date.
