Human Plasminogen Activator, Urokinase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of human Plasminogen Activator, Urokinase (uPA) in plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, where uPA binds to the immobilised capture antibody. A biotinylated anti-uPA antibody, HRP-avidin conjugate and TMB substrate are then introduced; the HRP reaction is read at 450 nm and the yellow colour intensity is proportional to uPA concentration, calculated against a standard curve. Sensitivity is reported as <7.1 pg/ml and the product range is 15.6-1000 pg/ml. Total assay time is 4h 30m.

Recovery data in serum (n=5) ranged from 80% to 102% (average 91%), in EDTA plasma (n=5) from 81% to 100% (average 90%), and in heparin plasma (n=5) from 80% to 89% (average 84%). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 is also provided for each sample type. Kit components include the pre-coated microplate, lyophilised standard (2 vials), detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or -20°C as indicated; the kit must not be used past the expiration date.
