Human Plasminogen Activator Inhibitor 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com for the in vitro quantitative determination of human Plasminogen Activator Inhibitor 2 (SerpinB2/PAI-2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for SerpinB2/PAI-2 pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells; following incubation and washing, a Biotinylated Anti-SerpinB2/PAI-2 Antibody is applied, followed by HRP-Avidin conjugate. After further incubation and washing, TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of SerpinB2/PAI-2 captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The product range is 0.156–10 ng/ml, with a sensitivity of 0.064 ng/ml. The assay is run on a pre-coated microplate (12 x 8 well strips) with a total assay time of 4h 30m. Kit contents include a Pre-Coated 96 Well Microplate, Lyophilized Standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and 5 Adhesive Plate Sealers. Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Reported recovery averages are 91% (range 80%–102%) in serum, 90% (range 81%–100%) in EDTA plasma, and 84% (range 80%–89%) in heparin plasma (n=5 for each). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5).
