Human Plasminogen Activator Inhibitor 1 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of human Plasminogen Activator Inhibitor 1 (PAI1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with enzymatic detection read colorimetrically at 450 nm in a microplate reader.

The kit reports a product range of 12.5–800 pg/ml and a sensitivity of <4.9 pg/ml, with a total assay time of 4 hours 30 minutes. An antibody specific for PAI1 is pre-coated onto the plate; standards and test samples are added and the bound PAI1 is detected using a biotinylated anti-PAI1 antibody followed by an HRP-avidin conjugate, with TMB substrate producing a yellow product whose absorbance is proportional to PAI1 concentration.

Reported mean recoveries are 91% in serum (range 80%–102%), 90% in EDTA plasma (range 81%–100%) and 84% in heparin plasma (range 80%–89%). The kit includes lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with components stored at +4°C or –20°C as instructed and not used past the expiration date.
