Human Plasmalemma Vesicle Associated Protein ELISA Kit

The Antibodies.com Human PLVAP/PV-1 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of human PLVAP/PV-1 in tissue homogenates, cell lysates, and other biological fluids. The assay employs a 96-well microtiter plate pre-coated with a PLVAP/PV-1-specific antibody. Standards and samples are added to the wells, followed by a biotinylated anti-PLVAP/PV-1 antibody and an HRP-avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue-coloured product that turns yellow after addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration of PLVAP/PV-1 is calculated against the standard curve.

Reported performance characteristics include an assay range of 0.156–10 ng/ml, a sensitivity of <0.059 ng/ml, and a total assay time of 4 h 30 min. Sample recovery was evaluated in serum (range 80–102%, average 91%, n=5), EDTA plasma (range 81–100%, average 90%, n=5), and heparin plasma (range 80–89%, average 84%, n=5). The pre-coated microplate is configured as 12 × 8 well strips, and kit components should be stored at +4 °C or −20 °C as instructed.
