Human Pim-1 Oncogene ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of human PIM1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is intended for use with a colorimetric microplate reader.

The kit employs the sandwich enzyme immunoassay technique using a pre-coated 96-well microplate. Standards and samples are added to wells where PIM1 is captured by an immobilised antibody, followed by incubation with a Biotinylated Anti-PIM1 Antibody, then HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of stop solution. The density of yellow is proportional to the amount of PIM1 captured, with concentration calculated by reading O.D. absorbance at 450 nm against a standard curve. The product range is 0.781–50 ng/ml with a sensitivity of <0.25 ng/ml, and total assay time is 4h 30m.

Validation studies (n=5) reported recovery ranges of 80%–102% for serum, 81%–100% for EDTA plasma, and 80%–89% for heparin plasma. Linearity testing across dilutions of 1:2 to 1:16 showed recovery values ranging from 74% to 107% for serum, 79% to 108% for EDTA plasma, and 82% to 105% for heparin plasma. Components are to be stored at +4°C or –20°C as instructed and must not be used past the stated expiration date.
